齿密螺旋体免疫抑制现象与T细胞凋亡的关系 被引量:1

摘要 用凝胶电泳DNA梯度图谱定性分析T淋巴细胞凋亡率;感染齿密螺旋体后不同时间的小鼠脾脏CD+4和CD+8T细胞,用流式细胞仪动态检测CD+4和CD+8T细胞的比值;两种细胞体外经齿密螺旋体超声破碎提取液刺激后,用流式细胞仪动态检测CD+4和CD+8T细胞的凋亡率.结果显示:(1)齿密螺旋体可诱导小鼠脾细胞凋亡;(2)实验组小鼠脾脏CD+4/CD+8T细胞的比值发生改变;(3)小鼠脾脏CD+4和CD+8T细胞于体外经齿密螺旋体超声破碎提取液刺激后,小鼠CD+4T细胞的凋亡率随时间的延长而明显增加;CD+8T细胞的凋亡率低下,且不随感染时间的延长而变化.通过对齿密螺旋体免疫抑制现象与T细胞凋亡的关系的研究表明:齿密螺旋体与宿主免疫功能减退、CD+4/CD+8T细胞的比值下降密切相关,其机制可能与齿密螺旋体超声破碎提取液含具有能引起T细胞凋亡的成分有关. The apoptosis rate of Tlymphocyte was analyzed qualitatively by gelatinizing electrophoresis DNA gradient atlas; the ratio between CD^+_4 and CD^+_8 T cells of mice spleen was checked by the flow-typed cell instrument in different time after injecting sonic extractus from treponea denticola; with sonic extractus from treponea denticola catalyzing the two kinds of cells, test the apoptosis rate of CD^+_4 and CD^+_8T cells by the flow-typed cell instrument. The Results shows that: (1) Treponea denticola can induce the mice spleen cells apoptosis; (2) The ratio of CD^+_4/CD^+_8T cells catalyzed by from treponea denticola came to be changeable; (3) With sonic extractus from treponea denticola catalyzing CD^+_4 and CD^+_8T cells of mice spleen in external, the apoptosis rate of CD^+_4T cells obviously increases with the extension of the time; but the apoptosis rate of CD^+_8T cells treaded to become lower, with no change by contracting time. The research of relationship between immuneinhibition of treponem denticola and apoptosis of T cells proves that Treponea denticola has close relation with immunologic decline of the host and the ratio decline between CD^+_4 and CD^+_8T cells, the mechanism of which concerns the components of sonic extractus from treponea denticola that cause the apoptosis of T cells.
