摘要
目的 探索层粘连蛋白促进兔角膜内皮细胞bcl 2表达的最适浓度。方法 给于体外培养的兔角膜内皮细胞以不同浓度的层粘连蛋白 ,采用酶联免疫技术找到最适作用浓度。结果 在一定的浓度范围内 (2 0~ 6 0mg·L-1)层粘连蛋白能够促进家兔角膜内皮细胞bcl 2的表达 ,并且成量效依赖关系 ,在 6 0mg·L-1时有最佳的促进作用。结论 层粘连蛋白可以促进兔角膜内皮细胞bcl 2基因表达的作用 ,最适作用浓度为 6 0mg·L-1。
Objective To investigate optimum concentration of laminin(LN)on the bcl-2 ex pression of rabbit's corneal endothelial cells. Methods Cultured rabbit's corneal endothelial cells were given LN of different concentrations and the ELISA was ch osen to measure OD.We compared the OD and got the optimum concentration of LN o f each group. Results Within certain concentration (20~60 mg·L -1 ), the LN could promote bcl-2 gene expression in rabbit's corneal endothelial cells in a dose- dependent manner with the optimal promote effect at 60 mg·L -1 . Conclusion LN could pr omote bcl-2 gene expression in rabbit's corneal endothelial cells,and its opti mal promote effect is at 60 mg·L -1 .
