摘要
目的 研究年龄相关性黄斑变性 (age relatedmaculardegeneration ,AMD)脉络膜新生血管膜基质金属蛋白酶 (matrixmetalloproteinases,MMPs)的表达 ,探讨MMP 2和MMP 9在AMD脉络膜新生血管膜形成过程中的作用。方法 实验组标本取自AMD患者经扁平部玻璃体切割术和视网膜下膜取出术获得的脉络膜新生血管膜 17例 ,免疫组织化学法检测MMP 2和MMP 9的表达。结果 17例AMD脉络膜新生血管膜MMP 2和MMP 9表达阳性 ,正常人视网膜无MMP 2和MMP 9表达。结论 AMD脉络膜新生血管膜有MMP 2和MMP 9的表达 ,MMP 2和MMP 9可能通过降解Bruch膜 ,促进脉络膜新生血管在视网膜色素上皮下生长 ,参与AMD的病理变化过程。
Objective To investigate the expression of matr ix metalloproteinases (MMPs) in choroidal neovascular membranes with age-related macular degeneration(AMD)and probe into the role of MMP-2 and MMP-9 in the for mation of choroidal neovascular membranes with AMD. Methods Seventeen choroidal neovascular membranes were obtained f rom AMD patients with pars plana vitrectomy and subretinal membranes peeling wer e investigated. The expression of MMP-2 and MMP-9 were determined with immunoh istochemical technique. Results Immunohistochemistry staining positive in choroidal neovascular membranes for MMP-2 and MMP-9 were observed in 17 cases with AMD.There was no detective of MMP-2 and MMP-9 in normal retinas. Conclusion MMP-2 and MMP-9 were found in choroidal neovascula r membranes,they may degrade the Bruch membrane and be associated with the perfo rat ion of new vessels into Bruch membrane,which involved a basic pathogenic process of AMD.
