摘要
本文用孔径为1200A的多孔玻璃珠作为载体,将乳酸脱氢酶固定于其上,装入柱内,联结于流动体系中。测定了固定化乳酸脱氢酶的表观米氏常数和人血清中的L-乳酸。L-乳酸的线性范围在0.1—9mmool/L,回收率为96—103%,变异系数<0.7%,最低检出限为0.4nmol(信噪比为4),每小时可进样35次,酶的稳定性良好。
The L-lactate dehydrogenase(LDH) is immobilized on macroporous glass beads(1200A) and packed into a column in the flow system. The apparant Michaelis constant of immobilized LDH and L-lactate in human serum are determined. The variant coefficient is <0.7%, the recovery rate 96-103% for the L-lactate concentration range of 0.1-9mmol/L, the detection limit 0.4nmol(S/N=4) and sampling frequency 35 samples/ h. The immobilized LDH is stable.
