高效液相色谱紫外检测氟哌酸血药浓度的方法

DETERMINATION OF NORFLOXACIN CONCENTRATIONS IN SERUM BY HIGH PRESSURE LIQUID CHROMATOGRAPHY

本实验用 LC-6A 高效液相色谱仪和 Spherisorb C18柱,在286 nm 波长处检测人血氟哌酸,用外标法峰面积定量(y=0.0410+0.000377x,r=0.9996,线性范围:0.249～4.97μg/ml)。回收率为83.9%～105.0%。日内和日间变异系数分别为2.74%～6.07%和1.02%～7.52%。用本法处理样品简单、出结果稳定迅速,适用于生物利用度试验和临床上氟哌酸血药浓度的测试。

A high pressure liquid chromatographic method for determination of norfloxacin concentra- tions in human serum was developed.A reverse—phase C18 column,an absolute calibration curve(slpoe, 0.000377; intercept, 0.0410;correlation coefficient,0.9996),and ultraviolet- detection were used.Interday and intraday variation coefficient ranged from 1.02% to 7.52% and 2.74% to 6.07% respectively.The absolute recovery of norfloxacin from serum over the concentration range of 0.5 to 5 μg/ml averaged 105.0% to 83.9%.
