应用放射性配基[H^3]-QNB分析法,观察10例病态窦房结综合征患者血淋巴细胞M受体的变化,淋巴细胞与[H^3]-QNB的特异性结合呈可饱和性,占总结合的70%;淋巴细胞M受体密度(B_(max)较正常对照组上升27%(P<0.05);而淋巴细胞M受体的亲和力(K_D)和正常组无显著差异。5例病窦患者窦房结恢复时间(SNRT)延长,与对应病例的B_(max)值增高呈正相关(r=0.92,P<0.05)。结果提示:血淋巴细胞上存在M受体;病窦患者血淋巴细胞M受体密度增加。
In order to explore the relationship between the disorder of sinus node and muscarinic receptor on human blood lymphocytes,the Bmax and Kd values of M-receptor on circulating intact lymphocytes were measured by[H^3]-QNB binding analysis in 10 patients with Sick Sinus Syndrome(SSS).Binding of[H^3]-QNB was linear with the increasing num- ber of lymphocytes and saturated in 8~10 nM of[H^3]-QNB.The Bmax values in patients with SSS were 27% larger than that in normal donors.There was a significant difference be- tween the Bmax values of normal blood donors and patients with SSS while no statistical dif- ference between the Kd values of them was found.Sinus node recovery time(SNRT)record- ed by esophageal atrial pacing was longer than normal range in 5 patients with SSS.The re- lationship between Bmax and SNRT from the same 5 individuals was well correlated(r= 0.92,P<0.05).These results indicate that alternation of M-receptor on circulating intact lymphocytes might be a linchpin in the process leading to disorder of sinus node.
