暴发性肝衰竭时脑内氨基酸类神经递质的研究

摘要 用致肝坏死药物 D-氨基半乳糖腹腔注射大鼠后测定额叶皮质,海马回,纹状体中GABA、谷氨酸、门冬氨酸、甘氨酸等氨基酸类神经递质的含量.结果揭示在暴发肝功能衰竭的状态下。无肝性脑病及有肝性脑病大鼠均表现为抑制性神经递质 GABA 在海马和纹状体的含量及甘氨酸在额叶皮质的含量增加;而在肝性脑病鼠海马中。兴奋性神经递质门冬氨酸含量减少.这表明氨基酸神经递质系统在肝性脑病发生中起重要作用.在高血氨状态下,静脉推注谷氨酸溶液(226mM,20ml/kg,bw/15min)。谷氨酸在脑中的含量增高.提示静脉推注谷氨酸溶液可使其通过血脑屏障,对肝性脑病有一定的治疗作用. Amino acid neurotransmitters,such as γ-aminobutyric acid(GABA),glutamate,aspartate and glycine were measured in the front cortex,hippocampus and striatum of rats with fulminant hepatic failure(FHF)induced with D-glactosamine.Whether the rats had hepatic encephalopathy(HE)or not,GABA,an inhibitory neurotransmitter,increased in hippocampus and striatum while glycine increased only in the front cortex.Aspartate,an excitory neurotransmitter,was reduced in the hippocampus of rats with HE.These foundings suggest that amino acid neurotransmitter system may play an important role in the course of HE. After a booster of glutamate(226mM.20ml/kg.bw/15m)intravenously applied to rats with HE and hyperammonemia,increased concentration of glutamate was detected in the brains,indicating that glutamate can cross blood-brain barrier and it may be of value in the treatment of HE.
