应用A蛋白分离纯化细胞培养上清中的鼠源性抗肥大细胞FcεRIα的单克隆抗体

Purification of monoclonal antibodies against mouse mast cell IgE receptor (FcεRIα) from culture supernatants by using protein A

目的 分离纯化鼠源性抗肥大细胞FcεRIα(高亲和力的IgE的Fc段受体)的单克隆抗体。方法 先培养ER-E5.3细胞,获得大量的含抗肥大细胞FcεRIα的单克隆抗体的培养上清。然后应用A蛋白-Sphrose CL-4B制备亲和层析柱,分离纯化细胞培养上清中的单克隆抗体。结果分离纯化的单克隆抗体纯度高,活性强。结论应用A蛋白可以很好分离纯化细胞培养上清中的鼠源性抗肥大细胞FcεRIα的单克隆抗体。

Objective To purify the monoclonal antibodies (McAbs) against mouse mast cell IgE receptor (FcεRIα). Methods A quantity of culture supernatant containing McAbs against FcεRIαwas obtained from the culture of ER - E5 .3 cells. The McAbs were then purified by chromatography with the high performance protein A - sepharose affinity column. Results The antibodies obtained were of high purity and activity. Conclusion Protein A - sepharose is a good means to purify the McAbs against FcεRIα.
