摘要
目的 观察人脑出血后其周围组织GFAP、Ki6 7在星形胶质细胞的表达 ,探索人脑出血周围组织星形胶质细胞活化和增生的机制。方法 采用HE染色和免疫组化染色 (两步法 )标记星形胶质细胞中的胶质纤维酸性蛋白 (GFAP)和Ki6 7,观察GFAP和Ki6 7在星形胶质细胞的表达和变化规律 ,实验结果应用SPSS11.5软件包进行统计学分析。结果 人脑出血后 8h其周围组织GFAP表达开始增加(P <0 .0 1) ,3~ 5d表达明显增强 (P <0 .0 1) ,7~ 15d表达达到高峰 (P <0 .0 1) ,以后有所回落 ,19d时仍高于对照组 (P<0 .0 1) ;Ki6 7阳性反应在脑出血 8h开始增强 (P <0 .0 1) ,3~ 5d数量增多 (P <0 .0 1) ,7~ 15d达高峰 (P <0 .0 1) ,随后表达呈下降趋势 ,19d时仍比对照组高 (P <0 .0 1) ,二者消长规律一致 (Pearsonr =0 .9881,P<0 .0 1,R2 =0 .976 3)。结论 星形胶质细胞在人脑出血后 15d内异常活化并与神经元的存活密切相关 ;脑出血性损伤使星形胶质细胞出现细胞周期的改变 ;Ki6 7参与了脑出血后星形胶质细胞的增生与活化。
Objective To observe expression of GFAP and Ki67,research the reactional mechanism of astrocytes in the surrounding areas after cerebral hemorrhage in human. Methods HE and immunohistochemistry were used to observe changes of glial fibrillary acidic protein (GFAP)and Ki67 in the surrounding areas after cerebral hemorrhage in human. Results The expressions of GFAP and Ki67 increased in 8h(P<0.01)and reached the peak in 7~15 days(P<0.01),their expressions had increased trend within 15 days after cerebral hemorrhage in human, after 15 days,the expressions of GFAP and Ki67 decreased and was higher than that in control group, the expression between GFAP and Ki67 showed positive correlation. (Pearson r =0.988 1,P<0.01, R 2=0.976 3)Conclusions Abnormally activated astrocytes are closely relative to neuronal survival within 15 days after cerebral hemorrhage;The cell cycle of astrocytes are changed after cerebral hemorrhage; expression of Ki67 leads to the activation and proliferation of astrocytes after cerebral hemorrhage.
