Apoptosis-related gene expression and its clinical significance of human osteosarcoma

摘要 目的 探讨反映骨肉瘤预后的蛋白标记物。方法 对骨肉瘤石蜡组织作免疫组化和Tunel染色 ,研究p5 3、c myc和bcl 2基因的表达与肿瘤细胞凋亡指数 (AI)的关系 ,及其与病理类型和患者预后的关系。结果 p5 3、c myc和bcl 2的表达水平与细胞凋亡指数呈负相关 ,与病理类型无相关性。p5 3、c myc、bcl 2和细胞凋亡指数与患者远期生存密切相关。结论 p5 3、c myc和bcl 2的表达水平以及细胞凋亡指数可作为判断骨肉瘤恶性程度和预后的指标 ,可指导临床进行治疗。 Objective To explore the prognostic markers in osteosarcoma. Methods Expressions of p53, c-myc, bcl-2 and apoptosis index (AI) in 28 osteosarcoma specimens were detected by ABC immunohistochemistry and terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-digoxigenin nick end labelling (TUNEL). The relationship between gene expression and apoptosis, and their correlations with pathologic classification and prognostic factors were analyzed. Results There was negative correlation between the expressions of p53, c-myc, bcl-2 protein and AI, which was closely related to the long term survival of patients but was not related to pathologic types of the tumor. Conclusion The expressions of p53, c-myc, bcl-2 protein and AI can be used as an index for predicting the progression and prognosis of osteosarcoma.
