摘要
目的 :评价色原对Trinder’方法测定微量葡萄糖的呈色效果。方法 :应用苯酚和 3种人工合成色原 :3,5 -二氯 - 2羟基苯磺酸钠 (色原 1)、N - 2 - [乙基 (3-甲基苯基 )氨基 ]乙基乙酰胺 (色原 2 )及N -乙基 -N -乙磺酸钠间甲苯胺 (色原 3)对葡萄糖氧化酶偶联过氧化物酶法测定微量葡萄糖的作用特点进行研究。结果 :使用 4种色原物质测定葡萄糖的反应曲线均类似 ,但呈色强度不同 ,苯酚呈色强度最差 ,色原 1最强 ,色原 2和 3类似 ;反应曲线为类似 0级反应模式 ;两点定标测定优于单点定标测定 ;批内不精密度范围 ,苯酚 :0 .79%~ 8.6 1% ,色原 1:0 .6 2 %~ 1.2 8% ,色原 2 :0 .83%~ 2 .6 0 % ,色原 3:0 .37%~ 3.80 % ,色原之间差异无统计学意义。结论 :在建立新的检测方法前要对色原进行评价。
Objective: To evaluate the colouring effects of chromogens in measuring micro-glucose with Trinder's method. Methods: The effective characteristics of phenol, 3,5-dichloro-2-hydroxide-sodium-benzenesulfonate(chromogen 1), N-[2-[ethyl (3-methylphenyl) amino]ethyl] acetamide(chromogen 2) and N-ethyl-N-ethyisodiumsulfonate 3-methylbenzenamine(chromogen 3)in testing micro-glucose with glucose oxidase coupling peroxidase were studied.Results:The reactive curves from four chromogens for testing glucose were similar. The colour intensitis were different. The chromogen one was most marked.Chromogen two and three were weaker,phenol was weakest. The reactive curve were o grade reactive mode. The measurement with double standards was better than that with single standard. The imprecision range was from 0.79% to 8.61% in phenol, from 0.62% to 1.28% in choromogen one, from 0.83% to 2.60% in chromogen two, and from 0.37% to 3.80% in chromogen three. There was no significant difference between choromogens. Conclusion: The colouring of chromogens should be evaluated before the new method is applied.
