摘要 建立了阿维菌素B1a在土壤生物蚯蚓体内的高效液相色谱荧光检测方法。匀浆后的蚯蚓样品用乙腈提取,经碱性氧化铝SPE柱和C18SPE柱净化,减压蒸干,与1-甲基咪唑(1-MIZ)和三氟醋酸酐(TFAA)衍生化反应后,进行HPLC荧光检测分析,激发波长为365nm,发射波长为475nm。添加法测定结果表明,当添加水平分别为5、50和500ng·g-1时,平均回收率分别为104.8%、86.6%和83.0%。变异系数在2.99%~7.34%之间。在蚯蚓组织中的检测限为0.5ng·g-1。 A method for determination of avermectin B1a in earthworms was established by high performance liquid chromatography (HPLC) with fluorescence detector. Homogenized earthworms sample was extracted by acetonitrile, and cleaned up on alumina B cartridge and C18 solid-phase cartridge, followed by derivatization with 1-methylimidazole (1-MIZ) and trifluoroacetic anhydride (TFAA), and analyzed by HPLC with fluorescence detector set at 365 nm excitation and 475 nm emission wavelengths. The limit of detection of the method is 0.5 ng·g-1. The average recoveries of avermectin were 104.8%, 86,6% and 83.0%, respectively in earthworm samples fortified at 5, 50, 500 ng·g-1 and the Coefficient of variation ranged from 2.99% to 7.34%.
