摘要
目的 探讨高压氧对缺氧缺血性脑损伤新生大鼠海马神经元线粒体的保护作用。方法 用新生 7日龄大鼠HIBD模型 ,在缺氧缺血后急性期给予 2 .0绝对大气压 (ATA)的高压氧 (HBO)治疗 ,每次 1h ,每日 1次 ,共 7d ,在治疗后 7d处死大鼠 ,用透射电镜观察受损侧脑组织海马神经元线粒体超微结构的改变 ,Western blot检测神经元胞浆细胞色素C含量的变化。结果 新生大鼠HIBD模型缺氧缺血后 7d神经元线粒体数量明显减少 ,形态异常 ,表现为线粒体体积缩小 ,嵴结构模糊、紊乱 ,有的呈现空泡样变 ,且神经元胞浆细胞色素C的含量升高 ;与HIBD组相比 ,HBO治疗后神经元线粒体的数量减少较HIBD组轻 ,线粒体的形态较规则 ,结构较完整和清晰 ,神经元胞浆细胞色素C的含量降低。结论 新生大鼠HIBD模型缺氧缺血后 7d线粒体的形态和结构有异常改变 ,从而使线粒体内的细胞色素C释放入胞浆 ,导致神经元胞浆细胞色素C的含量升高。HBO治疗后神经元线粒体数量的减少和超微结构的改变减轻 ,神经元胞浆细胞色素C的含量降低 ,其机制可能与HBO治疗改善了脑组织神经元的能量代谢 ,使线粒体因能量代谢障碍受到的损害减轻 。
Objective To explore the protective effect of HBO on the mitochondrion of newborn rat with hypoxic-ischemic brain damage(HIBD).Methods 36 newborn rats were divided into three groups randomly:normal, HIBD and HBO.After hypoxic-ischemic brain damage, the rats in HBO group were treated with 2.0 ATA HBO 1 hour daily for 7 days.After 7 days of the treatment, the ultrastructure of mitochondrion of hippocampus on newborn rats were observed by transmission electron microscope and the cytochrome c (CytC) in neuronal cell plasma were tested by western blot.Results The abnormal changes of the mitochondrion on newborn rats such as the decrease of mitochondrial number and the change of the mitochondrial shape and structure in HIBD group were observed and the quantity of CytC in neuronal cell plasma in HIBD group were higher.Compared with those of HIBD group, the degree of mitochondrial damage in HBO group was milder and the quantity of CytC were in neuronal cell plasma in HBO group was lower.Conclusion There were some ultrastructural changes of mitochondrion in HIBD group with a higher level of CytC in neuronal cell plasma.HBO can protect the mitochondrion from the hypoxic-ischemic brain damage, so the changes of the mitochondrion in HBO group were milder and the quantity of CytC were lower in HBO group.
