神经元τ蛋白过度磷酸化时14-3-3ξ的变化

摘要 为探讨τ(tau)蛋白异常磷酸化时神经元内 14 3 3ξ蛋白含量的变化 ,采用GF 10 92 0 3X和wortmannin联合注射大鼠侧脑室 ,诱导大鼠海马神经元τ蛋白发生过度磷酸化 ,同时观察 14 3 3ξ蛋白含量的改变 .结果显示 :GF 10 92 0 3X和wortmannin诱导了海马CA3区神经元τ蛋白在Ser396 / 4 0 4位点的过度磷酸化 ,同时14 3 3ξ蛋白的含量显著增高 .这些结果提示τ蛋白过度磷酸化与抗凋亡蛋白 14 3 3之间可能的内在联系 . To investigate the alteration of 14-3-3ξ during hyperphosphorylation of tau, hyperphosphorylation of tau was induced by injecting wortmannin and GF-109203X into lateral ventricle of rat. Phosphorylation of tau in hippocampal neurons and the alteration of 14-3-3ξ protein were detected. The results demonstrated that wortmannin and GF-109203X induced hyperphosphorylation of tau at Ser396/404 in CA3 region of hippocampus. The content of 14-3-3ξ protein increased. The results showed that a positive correlation between hyperphosphorylation of tau and 14-3-3 (an anti-apoptotic proteins).
