一种新的ELISA无毒底物TMBS在血清结核菌抗原检测中的应用

摘要 通过实验室操作,介绍3,3,5,5,一四甲基联苯胺硫酸盐(TMBS)作为ELISA底物,应用于血清结核抗原的检测;TMBS的主要优点是无致癌性,敏感性高,显色结果终止后稳定。通过实验确定了TMBS在ELISA中的最佳反应条件;经试用于Ⅲ例各种结核病人血清结核抗原检测,结果敏感性是目前最常用的底物邻苯二胺(OPD)的2.3倍,阳性检出率增加10.81%差异显著(P<0.05)。 3, 3', 5 , 5 '-ttetramethylbenzidine sulphate(TMBs), a new substrate of horseradish peroxidase (HRP) was used in ELISA for detecting M.tuberc. antigens in serum of 111 patients with one of three kinds of TB. TMBS't main advantages are noncarcinogenic, highly sensitive, and rather stable. Optimum conditions for the TMBS reaction were determined. Viaindirect ELISA, the sensitivity of TMBS was 2.3 times stronger than the conventionally used orthro-phenylene-diamine (OPD); the positive detecting rate wth TMBS was increased by 10.81% (p<0.05) . It seems that, the authors have the priority of utilizing TMBS in ELISA for detection of TB antigens.
