摘要 目的:了解嗅鞘细胞移植对脊髓损伤后神经细胞凋亡的影响,探讨嗅鞘细胞治疗脊髓损伤的机制。方法:成年动物脊髓半切损伤模型,进行嗅鞘细胞和培养液移植,在伤后1~14d,应用苏木精-伊红和TUNEL法观测神经细胞存活和细胞凋亡变化。结果:脊髓半切后,神经元数目随时间下降。伤侧和对侧分别在伤后1d和2d,出现以神经元为主的凋亡高峰。7d出现以胶质细胞为主的凋亡高峰,14d凋亡下降。嗅鞘细胞移植可明显降低脊髓损伤后神经细胞凋亡,促进神经细胞存活。结论:脊髓损伤后嗅鞘细胞移植有对抗神经细胞凋亡作用。 AIM:To investigate the effect of olfactory ensheathing cells(OECs) transplant on neuron apoptosis after spinal cord injury(SCI),and explore the mechanism of SCI treated with OECs. METHODS:Spinal cord hemisection was performed in adult animals. OECs and culture medium were transplanted.The ratio of surviving neurons and the change of apoptosis were observed with hematoxylin-eosin(HE) staining and TUNEL method at 1 to 14 days after operation. RESULTS:After spinal cord hemisection, the number of neurons in the spinal cord decreased with time.The peak of neuron apoptosis appeared on the 1st and 2nd day respectively after injury on the injured side and on the opposite side.Apoptosis of glial cells peaked at 7 days and declined at 14 days.OEC transplantation could reduce apoptosis of neurons after SCI and promote the survival rate of neurons. CONCLUSION:OEC transplantation after SCI has effect against apoptosis of nurons.
