摘要
目的:研究辣椒素对大鼠三叉神经节降钙素基因相关肽(calcitoningene-relatedpeptide,CGRP)免疫反应性神经元的影响,探讨辣椒素治疗三叉神经的镇痛机制。方法:把辣椒素配成浓度为30mmoL/L溶液备用,20只Wister大鼠按辣椒素用量分成4组(A组20μL,B组30μL,C组50μL和D组100μL),大鼠左侧面区为对照侧,右侧面为实验侧,分别皮下注射处理大鼠三叉神经眶下支,24h取材,切片,免疫组化染色,镜检,计算机图像分析。结果:经方差分析,对照侧,A,B,C,D组实验侧CGRP平均吸光度均值分别为2.526,2.499,2.199,1.812,1.549,组间比较差异有显著性意义(F=170.645,P<0.01)。并且随着用药量的增加其差值也更大。结论:辣椒素对大鼠三叉神经节CGRP免疫反应性神经元有明显的影响,CGRP参与口面部的痛与镇痛。
AIM:To investigate the influence of capsaicin on the calcitonin gene-related peptide(CGRP) immunoreative neurons in the trigeminal ganglia of rat,and probe into the analgesic mechanism of capsaicin in treating trifacial neuralgia. METHODS:Capsaicin was prepared into solution of 30 mmol/L for subcutaneous injection at infraorbital region to infiltrate the infraorbital branch of trigeminal nerve.The volume of solution injected for 20 Wistar rats was 20 μL(group A),30 μL(group B),50 mL(group C) and 100 μL(group D) respectively,and there were 5 rats in each group.Twenty-four hours after injection,the tissues of relevant region were sampled for immunohistochemical observation and computer image analysis. RESULTS:Analysis of variance showed that the mean absorbance(A) value of CGRP was 2.526,2.499,2.199,1.812 and 1.549 respectively in the groups A,B,C,and D,there were significant differences among the groups(F=170.645,P< 0.01),and the differences became greater with the increases of dosage. COUCLUSION:Capsaicin has an obvious effect on the CGRP immunoreative neurons in the trigeminal ganglia of rats,and CGRP participates in the mechanisms of pain and analgesia at oral-facial region of rats.
