猫儿眼植物酸对人HL-60细胞抑制作用的机制 被引量:15

摘要 目的:分离提取猫儿眼植物酸并探讨其对人HL-60细胞抑制作用机制。方法:实验于2003-07/2004-07在甘肃省医学科学研究院药理毒理学实验室完成。采用流式细胞仪、克隆形成和噻唑蓝(简称MTT)法对猫儿眼植物酸抗肿瘤细胞增殖作用进行分析。结果:分别给予人HL-60细胞猫儿眼植物酸0.1,1.0和10.0ng/L,MTT法测定,对人HL-60细胞增殖的抑制率分别为24.6%,69.6%,83.1%;克隆形成抑制率分别为29.3%,55.8%和71.4%;凋亡率分别为6.4%,8.8%和21.8%。结论:猫儿眼植物酸对人HL-60细胞增殖具有明显的抑制作用。 AIM:To separate and extract Maoeryan phytic acid,and then probe into the mechanism of its inhibitory effect on human HL-60 cells. METHODS:The experiment was completed in the Laboratory of Pharmacology and Toxicology,Gansu Academy of Medical Sciences from July 2003 to July 2004.The inhibitory effect of Maoeryan phytic acid on the proliferation of tumor cells were determined by flow cytometry,cloning and methyl-thiazolyl-tetrazolium(MTT) assessment. RESULTS:The Maoeryan phytic acid at a dose of 0.1,1.0 and 10.0 ng/L were given to human HL-60 cells respectively.After MTT assessment,the inhibitory rate for human HL-60 cells was 24.6%,69.6%and 83.1%respectively,the inhibitory rate for cloning was 29.3%,55.8%and 71.4%respectively,and the apoptosis rate was 6.4%,8.8%and 21.8%respectively. CONCLUSION:Maoeryan phytic acid has significant inhibitory effect on the proliferation of human HL-60 cells.
