高产菊粉酶的黑曲霉菌株产酶酶系分析与研究

摘要 对AspergillusnigerH8、A.nigerM89和突变株A.nigerUγ-2三株菊粉酶高产菌株产生的主要酶系进行了分析与研究。结果表明,在产菊粉酶的优化培养基(菊芋汁培养基)中进行摇瓶发酵,三菌株除产菊粉酶活力较高外,还产生蛋白酶,果胶酶,淀粉酶,纤维素酶;三株菌都表现为酶活力高峰随发酵时间依次是蛋白酶、淀粉酶、果胶酶和菊粉酶,纤维素因不同菌株有一定差异;突变株A.nigerUγ-2与其出发菌株A.nigerM89产酶比较表明,突变株在菊粉酶产酶活力获得大幅度提高的同时,淀粉酶和果胶酶的产酶活力也显著提高。 The enzyme systems of Aspergillus niger strains which have high inulinase activity were studied and analysed. The results show that the three strains could produce inulinase, proteinase, pectinase, amylase and cellulase, and the mutant strain A.niger Uγ-2 also had obviously amylase and pectinase activities and mutation prolong the production of cellulase. Compared with its initial strain A.niger M89, the mutant strain A. niger Uγ-2 showed not only higher inulinase but also amlyase and pectinase activities than its initial strain A.niger M89.
