摘要
应用反转录一聚合酶链式反应 (RTPCR)技术 ,从鸡新城疫I系疫苗诱导的鸡胚和成鸡脾脏及成鸡白细胞中提取的总RNA中扩增得到鸡白介素 2 (IL 2 )基因cDNA。结果表明 :以 5 0倍或 10 0倍稀释度接种鸡胚 4 8h提取脾脏总RNA扩增效果最好 ;以 5倍或 10倍免疫剂量免疫成鸡 4 8~ 72h ,提取脾细胞或血液白细胞总RNA扩增效果最佳。为进一步研究鸡IL
A cDNA encoding chicken interleukin(chIL2) was cloned from chicken and chicken embryo splenocytes as wall as white blood cells bw y reverse transcriptionpolymerase chain reaction (RTPCR)L .The results showed that the best factor for mRNA isolated from spleen cells ofchicken embryo was 48 h after inoculated with 50× or 100× diluted Newcastle disease Virus I strain,and with 5× or 10× times dosage inoculated chicken,after 48h to 72h isolated spleen cells and white blood cell extracted the total RNA for RTPCR.
