人胚肢芽、骨骼肌提取液的运动神经营养活性研究 被引量:1

摘要 目的:从人胚胎肢芽、骨骼肌中提取对运动神经元具有生物活性的神经营养因子。方法:用人胚胎肢芽、骨骼肌制备提取液,以成年Wistar大鼠骨骼肌提取液及生理盐水为对照,在切断单侧坐骨神经的Wistar乳鼠动物模型上,检测运动神经营养活性。结果:在损伤侧局部注射提取液,术后30天损伤侧腰髓运动神经元存活率:第2月人胚肢芽提取液组为73.7%;第4~5月胎儿骨骼肌提取液组为67.2%;成年大鼠骨骼肌提取液组为48.2%;生理盐水空白对照组为25%。结论:人胚胎肢芽、骨骼肌提取液与两个对照组相比均显示了明显的运动神经元营养活性。 Objective: To extract the neurotrophic factors with motonearon neurotrophic activity from the limb buds of embryo and the skeletal muscles of foetus. Methods: In the present study, the extracts were prepared from the limb buds of embryo, the skeletal muscles of foetus. The skeletal muscles extracts of adult rat and physiological saline were treated as controls. The motoneuron neurotrophic activities of those extracts were detected in sciatic nerve injured malels of neonatal wistar rats of 3 days age. Results: Thirty days following resection of sciatic nerve. by multiple injection of the second months embryo' s limb buds extracts at the site of axo-tomy, 73.7 % of the corresponding motoneurons were protected from cell death. The motoneurons survival rate was 67.2 % when treated with 4 - 5months foetus' s skeletal muscle estracts. and 48.2 % of adult rats' skeletal muscle extracts. In blank control (injecting physiological saline), there are only 25% of the corresponding montoneuron can survive. Conclusion:The result indicated that the extracts of embryo's limb buds and foetus'
