反复自然流产患者封闭抗体及CD_(25)分子的流式分析

Flow-type analysis of blocking antibody and CD25 molecule of patients with recurrent spontaneous abortion

目的探讨抗CD4,CD8,CD3和HLA-DR封闭抗体及淋巴细胞CD25在反复自然流产(RSA)诊治中的临床意义。方法应用流式细胞仪分别检测对照组及患者治疗前后淋巴细胞CD25及其配偶淋巴细胞与患者血清作用后的CD4,CD8,CD3和HLA-DR表达率。并通过与正常男性AB血清进行对照,计算出正常生育者或RSA患者血清对其配偶淋巴细胞CD4,CD8,CD3和HLA-DR的封闭效率。结果RSA患者CD4,CD8,CD3和HLA-DR封闭效率(BE,%)明显降低,依次为(2.5±1.4),(1.6±2.1),(8.1±3.9)和(2.6±3.7)。淋巴细胞CD25明显升高(6.4±1.9),与正常妊娠组及治疗后均差异有显著性(P<0.01)。结论抗CD4,CD8,CD3和HLA-DR的封闭抗体在正常妊娠中有重要的生物学作用,封闭抗体及淋巴细胞CD25的流式分析有可能成为RSA患者的辅助诊治和判断疗效的指标之一。

Objective: To explore the clinical significance of anti-CD4, CD8, CD3, HLA-DR and lymphocyte CD25 in the diagnosis and treatment of Recurrent Spontaneous Abortion (RSA). Methods: Lymphocyte CD25 of the patient group was detected with flow-type cyto-instrument before and after treatment as well as lymphocyte CD25 of the control group. The expression rates of CD4, CD8, CD3 and HLA-DR were detected after the lymphocytes of their spouses reacted to the serum of the patients. By contrasting with normal male AB serum, the blockyng efficiency of the serum of normal pregnancy or RSA patients to the lymphocyts CD4, CD8, CD3 and HLA-DR of thein spouses were calculated. Results: The blocking efficiency (BE,%) of CD4, CD8, CD3 and HLA-DR of RSA patients decreased notably, being respectively (2.5±1.4), (1.6±2.1), (8.1±3.9) and (2.6±3.7). Their lymphocyte CD25 rose notably (6.4±1.9), being significantly different from that of normal pregnancy group or the post-treatment value (P <0.01). Conclusions: The blocking antibody of anti-CD4, CD8, CD3 and HLA-DR has important biotic effect on normal pregnancy. The flow-type analysis of blocking antibody and lymphocyte CD25 may be used as a auxiliary index in the diagnosis, treatment and judgement of the rapeutic effect of RSA patients.
