不同切制条件对大黄饮片中蒽醌化合物含量的影响 被引量:7

摘要 目的 :采用高效液相色谱法比较不同的大黄切制条件对蒽醌类化合物含量的影响。方法 :以大黄素和大黄酚为对照品 ,采用MerckC18色谱柱 (4.6mm× 2 5 0mm ,5 μm) ,甲醇 -水 -磷酸 (体积比 78.7∶2 0 .9∶0 .4 )为流动相 ,检测波长 2 5 4nm ,外标法测定了不同切制条件下得到的 18份饮片样品中游离和结合型大黄素和大黄酚的含量。结果 :从 18份样品测定的平均值来看 ,游离型大黄素和大黄酚的含量为 8.6 9mg/g ,结合型的为 4 .4 5mg/g。从切制条件来看 ,薄片、中片、厚片总蒽醌 (即游离型和结合型大黄素和大黄酚之和 )含量平均分别为 13.4 7、13.33和 12 .6mg/g ;6 0℃干燥 10 .5h、80℃干燥 6 .5h和 10 0℃干燥 4h ,其总蒽醌含量平均分别为 13.73、12 .86和 12 .8mg/g。结论 :大黄类饮片切制的适宜条件为薄片或中片 ,6 0℃干燥 10 .5h。 Objective:Using HPLC methods to compare the contents of anthraquinones contained in the processed drugs of rhubarb obtained under different processed conditions. Methods:With emodin and chrysophanol as the reference substances,the separation was conducted on a column of Merck C18 (4.6 mm×250 mm,5 μm),using a mixture of methanol-water-H 3PO 4 (78.7∶20.9∶0.4) as the mobile phase and detected at 254 nm. Results:According to the average contents of the 18 samples,the sum of free emodin and chrysophanol is 8.69 mg/g,that of bonded emodin and chrysophanol is 4.45 mg/g. According to the precessing conditions,the average contents of total emodin and chrysophanol (including free and bonded) in thin piece type,medium piece type and thick piece type were 13.47、13.33 and 12.6 mg/g,respectively;those dried at 60 ℃ for 10.5 h,at 80 ℃ for 6.5 h and at 100 ℃ for 4 h were 13.73、12.86 and 12.8 mg/g. Conclusion:That the crude drug cut into thin or medium pieces and dried at 60 ℃ for 10.5 h is the appropriate condition for rhubarb processing.
