直肠癌相关新基因的荧光定量PCR检测 被引量:7

摘要 目的 应用实时荧光定量PCR技术检测人类新基因C1 0orf4 6ky(Genbank登录号为AY74 36 6 3)在直肠癌中表达情况及其意义。方法 制备31例直肠癌患者癌及癌旁正常组织中总RNA并逆转录为cDNA。应用实时荧光定量PCR技术分析C1 0orf4 6ky基因的mRNA表达水平。结果 在直肠癌癌组织中的C1 0orf4 6ky基因的mRNA表达水平高于相邻的癌旁正常直肠组织,但其表达水平与直肠癌的分期和病理分化程度在统计学上没有显著相关性。结论 人类新基因C1 0orf4 6ky在直肠癌中表达升高,暗示其与直肠癌变可能有关。 Objective To study the significance of the human's novel gene C10orf46ky in the rectum carcinoma by real-time fluorescence quantitative PCR.Methods The total RNA isolated from 31 example human rectum carcinoma and adjacent normal rectum tissue and was reversely transcribed into cDNA. The real-time fluorescence quantitative PCR method was used to analyze the expression level of C10orf46ky gene.Results The expression level of Cky10orf46ky gene was higher in tumor tissue than that in para-tumor rectum tissum. But it has no statistical significance between the expression level of human novel gene C10orf46ky and different stages of the rectum carcinoma and different degree of pathology of the rectum carcinoma.Conclusion The expression of human novel gene C10orf46ky up-regulate in the rectum carcinoma tissue. It imply that the up-regulation expression of human novel gene C10orf46ky may be related with rectum carcinoma.
