摘要
目的 建立测定阿齐霉素血药浓度的微生物法。方法 血药浓度采用微生物法测定。结果 阿齐霉素的标准曲线Y =0 .1136X - 4 .0 2 0 3(r=0 .9985 ) ;线性范围为 0 .0 5~ 0 .4 0 0 μg/ml,具有良好的线性关系 ;其 0 .0 0 5、0 .10 0、0 .4 0 0 μg/ml3个浓度的血清样本回收率分别为 10 0 .0 %± 0、10 7.8%± 5 .9%、10 3.7%± 6 .9% ;日内变异分别为 0、5 .5 0 %、6 .6 3% ;日间变异分别为 9.5 2 %、2 .5 3%、5 .2 0 %。结论 该方法简便快速 ,满足测定要求 ,可用于阿齐霉素的药代动力学研究。
Objective To set up a microbiological assay method for determining the concentration of azithromycin in serum.Methods The concentration of azithromycin was measured by microbiological assay.Results The standard curve of azithromycin in serum was Y=0.1136X-4.0203(r=0.9985). A good linearity was obtained from 0.05~0.400 μg/ml.The recoveries from concentrations of 0.005,0.100,and 0.400 μg/ml were 100.0%±0,107.8%±5.9%,and 103.7%±6.9%,respectively. The RSD of intra-day variation of azithromycin were 0%,5.50%,and 6.63% and the RSD of inter-day variation were 9.52%,2.53%,and 5.20%, respectively.Conclusion The method is simple,accurate and efficient for determining the concentration of azithromycin in serum and can be used to study pharmacokinetics of azithromycin.
