Expression of the ES Antigen Gene of Trichinella Spiralis

摘要 目的 完成旋毛虫 (Trichinellaspiralis,Ts)湖北株排泄分泌抗原 (excretory secretoryantigen ,ES抗原 )中 4 3ku ,4 9ku及 5 3ku抗原的不同虫期的基因表达 ,并与旋毛虫昆明株中相对应的ES抗原进行对比 ,分析不同虫期、虫株对这 3类抗原基因表达的影响。方法 根据已知的 3类抗原的基因序列设计合成 3对特异性引物 ,运用RT PCR技术检测抗原基因的表达 ,并以旋毛虫 18sRNA为标准进行对比。结果 RT PCR得到了湖北株两个虫期中的 4 3ku,4 9ku及 5 3ku的ES抗原mRNA产物 ,并与昆明株进行对比后发现 3类抗原在不同虫期表达有明显差异。结论 通过研究发现不同虫期的同类ES抗原的表达量有显著性差异。 Objective To study the 43 ku, 49 ku and 53 ku excretory and secretary (ES) antigens of Trichinella spiralis (Hubei isolates and Yunnan isolates) at pre-encysted stage and encysted stage, and analyze the expression of ES antigen gene at different development stage.Methods Three pairs of primer were designed according to the known sequence of ES antigen. Reverse transcriptase-PCR (RT-PCR) was used to identify the ES antigens of T S(Trichinella Spiralis).Results Electrophoresis results showed that the 43 ku and 49 ku antigen expressed at different stage, but the 53 ku antigen only expressed in encysted stage.Conclusion Different ES antigens have different expression level in specific stages.
