猪糖尿病皮肤溃疡模型建立的实验研究

Establishment of diabetic ulcer model in pig

目的:为适应糖尿病皮肤溃疡的临床和基础研究,建立一种简单而稳定的新型糖尿病皮肤溃疡模型。方法:选用约克猪6只,静脉注射四氧嘧啶,待空腹血糖达到1.2g/L以上,剃毛后在背部脊柱两侧安装自行研制的压创装置,分别于安装后6,12,24h后去除装置,根据压力和作用时间将创面分为10组,每组6个创面。不同时间点对创面进行大体观察、组织学检查以及创面面积测量。结果:对照组无溃疡发生;6h各组仅皮肤颜色改变,于去除装置一段时间后完全恢复正常;12,24h各组1周内均出现不同程度的溃疡,7～14d溃疡持续存在且无明显愈合趋势,14～21d溃疡收缩但未完全愈合。结论:该模型可模拟临床上糖尿病性皮肤溃疡且方法简便、重复性强。

AIM:To establish a new simple and steady diabetic ulcer model for the clinical and basic study of diabetic skin ulcer.<METHODS:Six York pig were injected alloxan through vein.The skin on back of the pig was shaved when the level of fasting blood glucose samples exceeded 1.2 g/L,and a kind of self designed press apparatus was installed on each side of spinal column.The press apparatus were respectively removed at 6,12 and 24 hours after installation.According to pressure and duration of apparatus acting on,the wounds were divided into ten groups,with six wounds in each group.The wounds was examined by gross and histological analysis, the area of wounds were measured. <RESULTS:No ulcer appeared in the control group.Only color changed in each group at 6 hours, and the color of wounds recovered completely some time after the apparatus was removed. Different degree ulcer occurred in each group at 12 hours and 24 hours on the 1st to 7th day after removal, and continuously existed without obvious sign of contracting on the 7th to 14th day, and the ulcers began to contract but did not heal completely on the 14th to 21st day.<CONCLUSION:The animal model can simulate clinical diabetic skin ulcer and the method of establishing such model is easy and reliable.
