高效液相色谱法测定子宫内膜细胞释放的前列腺素F_(2α)

摘要 目的:建立测定子宫内膜细胞在缩宫素(OT)存在的条件下分泌PGF2的方法。方法:采用柱α前衍生法对荧光衍生化试剂4-溴甲基-6,7-二甲氧基香豆素(BDMC)与PGF2的反应条件进行研α究。探讨荧光产物的色谱分离条件。结果:建立了测定子宫内膜分泌的微量PGF2的方法,测定了αOT诱导的大鼠子宫内膜细胞产生的PGF2。结论:本方法灵敏,最小检测限可达0.1ng,精密度α高,重复性强,可用于细胞水平或其他生物样品中前列腺素F2的测定。 Objective: To establish a method for determining the PGF2 in the endometrial cell which is α stimulated by oxytocin (OT). Methods: 4-bromomethyl-6,7-dimethoxycoumarin(BDMC)was used as a highly sensitive fluorescence derivative reagent. We investigate the reaction conditions of BDMC and PGF2 , discuss α isolation conditions of derivative products by high performance liquid chromatography. Result: We established the method for determing the trace PGF2 , and determined the PGF2 in the rats endometrial cell which is stimulated by α α OT. Conclusion: This method is sensitive, reliable, and could be used to determine the trace PGF2 α.
