摘要 目的 研究甲醛对V79细胞DNA的交联作用 ,探讨甲醛的遗传毒性及机制。方法 以紫外线照射作为标准断裂剂 ,用彗星试验检测不同浓度、不同暴露时间甲醛诱导的V79细胞DNA的交联作用。结果 除 75 μmol/L组外 ,在其它 4个浓度组、3个染毒时间点 ,甲醛均可引起显著的DNA交联作用 ,彗星尾长和彗星细胞率显著低于仅紫外线照射的对照组 (P <0 0 1) ,且彗星尾长具有明显剂量 -反应关系 (P <0 0 1)。细胞暴露在甲醛中 2、4h ,15 0、3 0 0、60 0 μmol/L浓度组彗星尾长和彗星细胞率显著低于暴露在 1h的 (P <0 0 1)。结论 甲醛是一种DNA交联剂 ,具有诱导V79细胞DNA交联的作用 。 Objective To study the effect of formaldehy de on DNA crosslinking of V79 Chinese hamster cells and to discuss the genotoxic e ffects and mechanism of formaldehyde. Methods UV was used as st andard DNA breaking agent DNA crosslinking of V79 Chinese hamster cells induced by different concentrations of formaldehyde and different exposure time were det ected with comet assay. Results Formaldehyde could cause DNA cr osslinking of V79 Chinese hamster cells. Except for the 75 μmol/L dose, the cro sslinking were increased with concentrations of formaldehyde and expose time in vitro studies. Conclusion Formaldehyde was a DNA crosslin-king a gent that could cause DNA crosslinking in V79 Chinese hamster cells and DNA cros sl inking may be a biomarker detecting the effect of formaldehyde on the bio-body.
