胰岛素微乳大鼠结肠给药的药效学研究 被引量:12

摘要 目的建立胰岛素(Ins)大鼠结肠给药实验动物模型,考察胰岛素微乳对高血糖大鼠的药效指标。方法用链脲佐菌素制备大鼠高血糖模型,并制备了结肠给药大鼠动物模型,分别设立胰岛素生理盐水溶液组(阴性对照组),空白O/W微乳组(阴性对照组),5IU/kg胰岛素皮下注射组(阳性对照组),胰岛素微乳低剂量组(5/3IU/kg),中剂量组(5IU/kg),高剂量组(15IU/kg),考察高血糖大鼠结肠给药后的降糖效果。结果用链脲佐菌素制成的高血糖模型稳定可靠,一次性ip60mg/kg,5~7d后可形成稳定的高血糖模型;胰岛素微乳高、中、低剂量组经大鼠结肠给药后均能降低大鼠血糖浓度,并呈剂量依从性,15,5,5/3IU/kg三剂量使其血糖降低的最大百分率分别为614%,445%和281%。结论所建立的大鼠结肠给药实验动物模型能够较好地反应药物的疗效,胰岛素微乳可有效降低高血糖大鼠的血糖。 AIM:To investigate the efficacy of insulin microemulsion on the hyperglycemia rats and to build the animal model of colon administration.MEHTOD:Streptozotocin is used to build the hyperglycemia model of rats and the rats model of colon administration is also built.[Rats were divided into six groups:normal saline solution of insulin(negative control group),the blank O/W microemulsion group(negative control group),subcutaneous injection group of 5 IU/kg insulin(positive control group),low dose of insulin microemulsion group(5/3 IU/kg),medium dose group(5 IU/kg) and high dose group(15 IU/kg),and after respectively administration,the effect of decreasing blood sugar was investigated.]RESULT:The stable hyperglycemia rats model will be built after ip of 60 mg/kg streptozotocin and about 5~7 days;The high,medium and low dose group of insulin microemulsion could all decrease blood sugar of rats after colon administration and the dose-dependence could be found:the greatest percentage of lowering blood sugar is 61.4%,44.5% and 28.1%,respectively.CONCLUSION:The animal model of colon administration can reflect the effect of insulin microemulsion and insulin microemulsion can decrease blood sugar of rats effectively.
