摘要
为研究宽叶缬草提取物抗癫痫作用及其机理 ,用戊四氮 37 5mg/kg腹腔注射Wistar大鼠 2 8d建立慢性癫痫模型 ,同时胃管给予宽叶缬草提取物 (5 0 0mg/kg、10 0 0mg/kg、15 0 0mg/kg)每天 3次 ,观察大鼠行为及脑电图变化 ,用RT PCR检测GABA转运体 (GAT 1)mRNA的表达。结果 :大鼠 2 8d点燃后 ,GAT 1mRNA表达明显增加 ,与正常组比较 ,P <0 0 5 ;治疗组与模型对照组比较 ,P <0 0 5 ,大鼠癫痫发作次数明显减少 ,且发作时间缩短。提示 ,宽叶缬草提取物能有效对抗戊四氮的点燃作用 ,且有剂量依赖性 ,其机理可能是通过抑制GAT 1mRNA表达而起作用。
To investigate the effects and mechanism of Valeriana officinalis Var. latifolia (VOL) on pentylenetetrazol (PTZ) chemical kindling rats, a kindling model of chronic epilepsy in rats were established by the injection of PTZ intraperitoneally. RT-PCR was used to determine the expression of GABA transporter-1 (GAT-1) mRNA in the hippocampus tissue, and the effect of VOL on behavior was observed and EEG of hippocampus in epileptic rats was examined. Results: The GAT-1 mRNA expression of rats injected with PTZ was higher than that of normal rats (P<0.05); epilepsy attacked obviously less and lasted shorter period. It is indicated that VOL can fight against PTZ to inhibit GAT-1 mRNA, with dose independence.
