通脉宁对血管狭窄模型家兔病理学及相关因子影响

摘要 目的:观察通脉宁胶囊(TMNJN)对家兔颈动脉内皮球囊损伤后引起血管狭窄模型的预防治疗作用。方法:在无菌条件下分离出颈内和颈外动脉,由颈外动脉向颈内动脉插入气囊,在恒定压力下,反复抽拉3次,造成经腔内皮损伤,连续给药21天后取出颈内动脉,肉眼和苏木素伊红染色观察动脉壁外观和病理学改变;颈总动脉取血,分别检测血a-颗粒膜蛋白(GMP-140)、内皮素1(ET-1)、降钙素基因相关肽(CGRP)的变化。结果:家兔颈动脉内皮球囊损伤21天后模型组家兔颈内动脉内皮明显粗糙,管腔明显狭窄,狭窄处87.5%可见黄色或暗红色血栓,内膜明显增厚,平滑肌细胞明显增生;血浆GMP-140、ET-1、CGRP明显高于假手术组。造型家兔灌服通脉宁胶囊后,大、中、小剂量组上述指标均有不同程度的改善,特别是大剂量组改善明显,差异显著。结论:大、中剂量组家兔灌服通脉宁胶囊后对动脉球囊损伤后引起血管狭窄模型有较好的预防治疗作用,为通脉宁胶囊能治疗经皮经腔内皮损伤引起的血管狭窄提供了充实的药理学基础。 Objective :To observe effects of prevention and therapy of TMNJN after internal carotid artery balloon injury in rabbits models of vascular stenosis. Methods : With asepsis operation, internal and external carotid artery was separated, then the balloon was only inserted from external carotid artery to internal carotid artery, and draw back and forth repeatedly for three times in constant pressure, to result in carotid artery endodermis injury from lumen and vascular stenosis. After the dosus were given successively for 21 days, the blood was taken from the common carotid artery, ando-GMP-14, ET- 1, CGRP in plasma were respectively measured. Results :The internal carotid artery endodermis of rabbits models of 21 days after internal carotid artery balloon injury appeared significantly coarse, and vascular lumen was significantly strait. The yellow or dark - red thrombus can be identified in 87.5 % of stenosis parts, and endomembrane was significantly incrassated, and smooth muscle was significantly hyperplastic. Compared with the sham operation group, the content of GMP - 140, ET - 1, CGRP in plasma was higher significantly. Compared with model group, the above- mentioned indexes in TMNJN large and middle and small dose group, improved to some extent, especially large dose group improved significantly. Conclusion : TMNJN with large and middle dose has certain effects of prevention and therapy on rabbit model of vascular stenosis induced by vascular endodermis injury. Thus this study provides substantial pharmacological basis about therapy of vascular stenosis induced by vascular endodermis injury.
