棉花半胱氨酸蛋白酶基因的克隆和表达特性分析 被引量:6

摘要 根据不同植物编码半胱氨酸蛋白酶基因的保守序列,设计兼并引物,通过PCR和RACE技术,从辽棉9号衰老叶片中克隆了编码半胱氨酸蛋白酶的cDNA,其相应的基因命名为Ghcysp,该基因序列的长度为1368bp,包含一个1034bp的可读框,编码344个氯基酸,分子量为37.88kD,等电点为4.80,数据分析结果显示,该蛋白质由1个19个氨基酸残基组成的信号肽和3个酶活性催化反应中心组成,是一个跨膜蛋白质,位于液胞摸上,Ghcysp蛋白质与其他植物半胱氨酸蛋白酶的氯基酸同源性为67%~82%,Nonhem杂交结果表明,在棉花衰老的根、脱落的花、下胚轴、胚珠和幼叶中,Ghcysp基因不表达,在棉花的衰老叶片中Ghcysp基因表达量高。
