脱细胞保鲜异种骨移植免疫反应的实验研究

Immune reaction to transplantation of PBCCB in rats

目的 研究脱细胞保鲜异种骨移植的免疫排斥反应。方法 以大鼠为动物模型 ,分别植入脱细胞保鲜牛皮质骨(PBCTB)和松质骨 (PBCCB)、新鲜骨 (FBC)、去抗原牛松质骨 (BCB) ,采用体外二次刺激增殖实验 ,酶联免疫吸附实验及局部组织学观察 ,研究移植后大鼠体内的细胞免疫、体液免疫及宿主对该移植物的局部病理反应。结果 保鲜牛皮质骨、松质骨显著降低了异种骨的抗原性、其中保鲜松质骨尤佳 ,与BCB相比较 ,没有统计学差异。结论 脱细胞处理的保鲜松质骨显著降低了细胞的免疫原性 ,同时可以避免化学毒物的残留 ,是一种理想的植骨材料。

Objective To study immunological rejection to preserved bovine cancellous bone(PBCCB), preserved bovine cortical bone(PBCTB), fresh bovine cancellous bone (FBC) and antigen-extracted bovine cancellous bone(BCB). Methods Five groups of Wistar rats were implanted with PBCCB, PBCTB, FBC,BCB and nothing respectively. Antigen stimulated T-lymphocyte proliferation in vitro and levels of specific serum IgG using enzyme-linked immuosorbent assay and histological changes were detected to evaluate the levels of cellular, humoral immunity and tissue reaction. Results Rates of T-lymphocytes transformation and levels of specific serum IgG were higher in FBC group than in PBCCB, PBCTB, FBC and BCB groups. There was no difference between PBCCB and BCB groups in the immunological response. Conclusion PBCCB is a good material for bone xenotransplantation
