摘要
乙酰螺旋霉素(Ac-SPM)是近年来广泛应用于临床的一种十六碳大环内酯类抗生素,其体内外抗菌活性均优于螺旋霉素。Ac-SPM是双乙酰螺旋霉素Ⅱ、Ⅲ和单乙酰螺旋霉素Ⅱ、Ⅲ四个组分的混合物。我们采用柱色谱法,在薄层色谱法(TLC)的监测下,分离制得Ac-SPM的各组分标准品。
An HPLC method has been developed for the determination of Acetyl-spiramycin (Ac-SPM) in rabbit plasma. The method involves separation and quantification of four components of Ac-SPM. The compounds were separated on a Spherisorb column (4- 6 × 200mm, 5μm particle size) using a mobile phase of methanol/0. 01 mol/L phosphate buffer (pH 2- 8) (44 : 55). UV detection was performed at 232nm, with a limit of detection of 1. 00 mol/L. The intra- and inter-assay precision were≤3. 67% and≤2. 40%. The method was .successfully applied to plasma samples obtained from rabbits after oral administration of Ac-SPM.
