摘要 在缺氧和缺血性疾病中,机体能量代谢障碍的程度可用组织中高能核苷酸及其代谢物的含量变化来反映。3-硝基丙酸(3-NPA)是引起变质甘蔗中毒的主要毒素,它可抑制参与三羧酸循环的琥珀酸脱氢酶,使有氧氧化受阻而导致能量代谢障碍。本文参照Hull-Ryde方法建立了一个重复性好、灵敏度高的高能核苷酸及其代谢物的反相高效液相色谱测定方法。 A method for the determination of high energy nucleotide and its metabolites by RP-HPLC with Ultrasphere?C18 column at 254nm was established to study the change of energy metabolites in cerebrum of rats intoxicated with 3-nitropropionic acid. The mobile phase A is 11. 5g NH4H2PO4 dissolved in 1000ml distilled water. The mobile phase B consisted of 11. 5g NH4H2PO4 and 200ml methanol dissolved in 800ml distilled water. This method was reproducible with CV range of 0.66-8.45%, and suitable to investigating changes of energy metabolites in several tissues besides brain.
