摘要
目的 确定多聚乙烯亚胺最佳的转染条件 ,评价PEI替代其它转染试剂的可能性。方法 以MTT法检测PEI的细胞毒性。以PEI为载体 ,在不同的N P比值的条件下 ,将绿色荧光蛋白基因和荧光素酶基因分别导入小鼠骨肉瘤细胞 ,以荧光显微镜酶检测系统检测其转染效率。结果 PEI的细胞毒性较低。当N P比值为 8时 ,PEI的转染效率最高。结论 PEI是一种价格低廉、低毒性、高效率的基因转染载体 ,可以广泛应用于体内外基因治疗。
Objective To investigate the best cond ition of polyethylenimine (P EI) for gene transfer to mouse osteosarcoma cells. Methods E valuate the cytoto xicity of PEI by Mononuclear cell direc cytotoxicity assay (MTT). In different c onditions, using PEI as the vector, mouse osteosarcoma cells were transfected wi th fluorescent protein (GFP) gene and luciferase gene. Expression efficiency of the transduced gene was monitored by fluorescence miceoscopy and luciferase assa y system. Results PEI had low cytotoxicity. When N/P ratio e qualed to 8, the t ransfection efficiency of PEI was highest. Conclusions PEI i s an inexpensive, low cytotoxicity and high efficiency vector. It can be widely used in the area o f gene therapy.
