摘要
目的 建立一种人 鼠异基因移植物抗宿主动物模型 ,为研究人 鼠嵌合抗体在动物体内移植排斥反应中的作用提供动物模型。方法 将人外周血单个核细胞移植到裸鼠体内 ,记录动物的存活时间 ;用流式细胞术对裸鼠脾细胞表型进行分析 ,并经组织病理学切片鉴定移植物抗宿主动物模型 (人外周血 裸鼠 )的建立。结果 外周血 裸鼠存活时间为 (2 2 5 0± 6 36 )d ;流式细胞术检测显示人外周血 裸鼠脾脏内人白细胞占 6 0 % ;组织病理学切片显示其肝脏、肺、脾脏有大量淋巴细胞浸润。结论 人 鼠异基因移植物抗宿主动物模型复制成功。
Objective To construct a mice model for xenogeneic graft versus host disease for studying the effect of mouse/human chimeric antibody (D2C) in anti-transplant rejection in vivo. Methods The naked mice engrafted with human PBMC as a model for xenogeneic graft versus host disease (XGVHD) were identified by FACS on cell su rface phenotype of human lymphocytes in its spleen and histological analysis of its tissues. Mice survivals of Hu-PBMC naked mice were recorded. Results Mice survivals of Hu-PBMC naked mice were (22.50±6. 3) days. FACS revealed that the spleens of Hu-PBMC naked mice were constituentl y engrafted with approximately 60 % human leukocytes. Histological analysis show ed massive T cell infiltration in liver, lungs, and spleen. Conclusion The mice model for XGVHD was successfully constructe d.
