色甘酸二钠抗大鼠肝纤维化作用的实验研究

摘要 目的 研究整合素α4β1及其 2个天然配体血管细胞粘附分子 1(VCAM 1)和纤维连接蛋白 (FN)在大鼠实验性肝纤维化时肥大细胞 (MC)募集过程中的意义以及色甘酸二钠 (DC)对防治肝纤维化的可能作用。方法 用 80只雄性SD大鼠制作CCl4诱导的肝纤维化模型 ,在光镜和电镜下观察MC与纤维组织的关系 ,应用免疫组化检测肝组织中整合素α4β1、VCAM 1及FN的表达。其中用 2 0只在造模前 30min给予腹腔注射DC 2 0mg/kg ,并与模型组、正常组的Ⅰ型及Ⅲ型胶原对比。结果 ①随着肝纤维化进展 ,大鼠肝组织中MC数量相应增加 ;② 76 4 %MCs呈整合素α4β1阳性表达 ;③肝组织中血管内皮细胞和肝窦内皮细胞VCAM 1呈阳性表达 ;④汇管区和小叶间隔内沉积大量阳性表达的FN。⑤DC组与模型组Ⅰ型及Ⅲ型胶原的表达差异有显著性意义 ,P <0 0 5。结论 整合素α4β1及其 2个配体VCAM 1和FN是大鼠肝纤维化过程中MC募集的重要因子 ;MC与肝纤维化有着密切的关系。DC在预防肝纤维化发生及其进展中有一定的作用。 Objective To study the role of intergrinα 4β 1/VCAM-1 and FN in mediating mast cell ( MC ) recruitment during rat liver fibrosis and th e probable role of disodium chromoglycate (DC) in preventing and treating rat li ver fibrosis. Methods Liver fibrosis models of 80 male SD rats were induced b y CCl 4. The relationship between MC and fibrous tissues was observed under lig ht and electron microscopy. Results ① The number of MCs in the liver tissue was increased correspondingly with fibrosis development. Most MCs distributed along collagenou s fibrils. ② 76.4 % MCs expressed integrinα 4β 1 positively. ③ Vascular e ndothelium in liver tissues and the hepatic sinusoid endothelium expressed VCAM -1 positively. ④ Largely expressed FN deposited between hepatic lobuli and in the periportal spaces. ⑤ There was significant difference in the expression of collagen Ⅰ and collagen Ⅲ between DC group and model group (P<0.05). Conclusion Ingtegrinα 4β 1 and its two ligands (VCAM-1 and FN) were very important pathways mediating MCs recruitment in rat liver fibrosi s. MCs were closely correlated to liver fibrosis. DC has some effect in preventi ng and treating rat liver fibrosis.
