摘要
为了解EHFV的抗原结构、基因突变,为EHF临床分型和疫苗生产提供资料。作者将EHFV标准株A9,R22与鲁宁-84刘、鲁宁-84肖、R-36的结构蛋白经SDS-PAGE分离,按改良氯氨T法用^(125)I标记凝胶中的NP,G_2和G_1,酶解后进行肽图分析。分别比较姬鼠型和家鼠型EHFV的NP、G_2、G_1肽图。结果发现各型病毒间有显著不同,氨基酸组成有明显差别,说明不同型别EHFV的S,M基因片段之间存在着差异。
In order to find out antigen structure,gene mutation,and
classification of epidemic hemorrhagic fever virus(EHFV)and supply some
information for vaccine production,we carried out SDS-PAGE to separate
the structural proteins of five strains.
The structural proteins of 5 EHFV strains were separated by SDS-
PAGE.NP,G_2,and G_1 were labelled with ^(125)I by a modification of the
chloramine T method and hydrolyzed by by trypsin.Tryptic peptide maps
showed that there were differences between apodemus type and house rat
type EHFV.The results indicated that remarkable differences existed between
amino acid sequences of these two kind of EHFV structural proteins.It was
also suggested that there were differences between the coding gene segments
(S and M)of these two kinds of EHFV.
