培养条件对黄孢原毛平革菌 Phanerochate chrysosporium ME—446 降解稻草木素、纤维素和半纤维素的影响

摘要 在固体培养条件下研究了培养条件对黄孢原毛平革菌Phanerochatechrysosporium ME—446降解稻草木素、纤维素和半纤维素的影响.试验采用正交设计.结果表明,ME—446菌株优先降解半纤维素.添加谷氨酸(3×10^(-6)mol/g草)可显著促进木素降解;基本营养盐显著促进半纤维素降解,但它们对纤维素降解均无显著影响;继续改变它们的用量并不能增大降解.高浓度的葡萄糖(5.5×10^(-4)mol/g草)极显著地抑制纤维素降解.醋酸缓冲夜pH4.5的缓冲强度对降解影响复杂.适宜的培养条件可以在获得有效木素降解的同时大幅度地减少纤维素损失。 Effects of culture conditions on lignin, cellulose and hemicellulose degradation of rice straw by Phanerochate chrysosporium ME-446 under solid-state fermentation was stud- ied. Orthogonal design was employed in experiments. The results showed, ME-446 strain degradates hemicellulose prior to lignin and cellulose. Lignin degradation is significantly increased by glutamic acid added (3μmol/g straw), and hemicellulose degradation is significantly increased by basal nutritional salts. But either of them significantly affects cellulose degradation. Degradation is not increased by continued raising concentration of either. High concentration of glucose (550μmol/g straw)can significantly suppress cellulose degradation.Capacity of acetate buffer pH4.5 has complicated effects on rice straw degradation. Effective lignin degradation could be obtained while loss of cellulose minimizes by adopting suitable culture condition.
