藻酸双酯钠对大鼠实验性急性脑缺血再灌注后p53表达的影响 被引量:7

THE EFFECT OF PSS ON p53 EXPRESSION AFTER REPERFUSION FOLLOWING RATS ACUTE CEREBRAL ISCHEMIC

摘要 目的 探讨大鼠急性脑缺血再灌注后 p53的表达及藻酸双酯钠 (PSS)对p53表达的影响。 方法 采用线栓法制作大鼠大脑中动脉阻断 (MCAO)再灌注模型 ,用免疫组化法分别检测缺血组和PSS治疗组大鼠脑缺血 2h ,再灌注后 1 ,3 ,6 ,1 2 ,2 4 ,48,72hp53蛋白的表达水平。结果 大鼠脑缺血 2h再灌注 1h皮质区可见 p53蛋白阳性细胞 ,2 4h达高峰 ,48h开始下降。与缺血组相比 ,PSS治疗组p53蛋白阳性细胞于再灌注后 6 ,1 2 ,2 4 ,48,72h表达均有减弱 (P <0 .0 5 ,P <0 .0 1 )。 结论 脑缺血再灌注后 p53蛋白表达增加 ,是脑缺血后的一个损伤因子 ;PSS可通过抑制p53蛋白的表达而对神经细胞具有保护作用。 Objective To study p53 expression after reperfusion following rats acute cerebral ischemic and the effect of PSS on p53 expression after reperfusion following rats acute cerebral ischemic. Methods We made middle cerebral artery occlusion(MCAO) and reperfusion model using an intraluminal filament method, and detected expression of p53 after 1,3,6,12,24,48,72h reperfusion following 2h cerebral ischemic in rats using method of immunohistochemistry in both ischemic and PSS treat groups. Results p53 protein positive cells were found in the cortex after 1h reperfusion following 2h cerebral ischemic in rats, reached peak after 24h, and reduced after 48h. Compared with ischemic group, p53 protein positive cells expression of PSS treat group weakened after 6,12,24,48 and 72h after reperfusion (P<0.05,P<0.01). Conclusion The expression of p53 protein increases after reperfusion following ischemic which is a damage factor after cerebral ischemic,and PSS can protect neuron by inhibiting p53 protein expression.
