EB病毒感染与子宫颈鳞状细胞癌相关性的研究 被引量:1

摘要 目的 探讨EB病毒 (epstein -barrvirus ,EBV)潜伏感染与子宫颈鳞状细胞癌 (cervicalsqua mouscellcarcinoma,CSCC)的关系。方法 采用免疫组织化学S -P法对 6例子宫颈上皮内瘤病变 (cervicalin traepithelialneoplasia,CIN)、 4 1例子宫颈鳞状细胞癌 (cervicalsquamouscellcarcinoma ,CSCC) (2 2例深圳 ,19例武汉 )和 1例子宫颈腺鳞癌进行单克隆抗体潜伏膜蛋白 1(latentmembraneprotein 1,LMP1)、EB病毒核抗原 2 (ep stein -barrviralnuclearantigen 2 ,EBNA2 )和EB病毒早期反式激活蛋白 (ZEBreplicationactivatorprotein ,ZEBRA)标记。对 36例CSCC组织进行EBERs原位杂交 (ISH)检测。结果 LMP1阳性部位位于癌细胞的胞核中。对照组阳性率 6 3% (1/ 16 ) ,CIN阳性率 33 3% (4/ 18) ,CSCC阳性表达率为 5 4 8% (2 3/ 4 2 ) ,三组间比较差异有显著性 (P =0 0 0 )。EBNA2仅在个别CSCC组织中有表达 ,余均为阴性。在所有研究标本中均未见ZEBRA表达。E BERsISH检测 36例CSCC阳性率为 38 9% (14 / 36 ) ,CSCCⅠ期阳性率为 4 0 0 % (2 / 5 ) ,CSCCⅡ期 (40 9% )与CSCCⅢ期 (2 7 3% )EBERsISH阳性率比较差异无显著性意义 (P =0 74 )。EBERs阳性信号均为散在灶状或单个癌细胞胞核阳性。结论 本文结果表明EB? Objective To study the correlation of Epstein-Barr virus (EBV) Latent infection with the human cervical squamous cell carcinoma (CSCC).Methods Six cases of cervical intraepithelial neoplasia (CIN), 41 CSCC (22 from Shenzhen,19 from Wuhan )and 1 adenosquamous carcinoma were examined immunohistochemically for latent membrane protein 1(LMP1), Epstein-Barr viral nuclear antigen2(EBNA2) and ZEB replication activator protein (ZEBRA).36 CSCC(17 in Shenzhen women and 19 in Wuhan ) underwent EBERs test by RNA in situ hybridization assays. Results LMP1 positive rate was 22.2%(4/18)in CIN,6.3% in normal cervix and 54.8%(23/42) in all of CSCC cases,There were significant difference between three gropus.(P=0.00). The positive rates of EBERs in CSCC cases were 38.9% (14/36). The EBERs positive signals scattered in CSCC nuclei. Conclusions EBV Latent infection may contribute to pathogenesis of CSCC, but does not play a major role in the etiology of CSCC.
