摘要
目的 探讨积雪草苷 (asiaticoside)在烧伤创面愈合过程中对细胞周期蛋白B1(CyclinB1)、细胞周期蛋白C(CyclinC)及增殖细胞核抗原 (PCNA)表达的影响。方法 建立Waster大鼠深Ⅱ°自身对照烫伤模型 ,分为积雪草苷治疗组和对照组。采用免疫组织化学方法 ,检测创面愈合过程中CyclinB1、CyclinC及PCNA表达水平。 结果 积雪草苷组在伤后第 3天开始有CyclinB1表达于基底细胞层。第 5天表达明显高于对照组 (P <0 .0 1)。PCNA免疫组织化学结果与CyclinB1表达一致 ,积雪草苷组在伤后第 5天表达明显高于对照组 (P <0 .0 1)。而CyclinC在积雪草苷治疗组和对照组于伤后第 4天胞核表达明显增多 ,但两组差异无统计学意义 (P >0 .0 5 )。结论 积雪草苷能有效促进CyclinB1和PCNA的表达 ,使细胞周期的S +G2 期明显提前 ,从而加快细胞增殖 ,促进创面愈合。
Objective To investigate the influence of asiaticoside on the expression of CyclinB1,CyclinC,proliferating cell nuclear antigen in healing tissue of burned rat.Methods The self control deepⅡ degree scalding models of Waster rats were established and immunohistochemistry method was employed to detect the expression of CyclinB1,CyclinC,PCNA in asiaticoside treatment group and in control group.Results The expression of CyclinB1 and PCNA in asiaticoside treatment group was significantly higher than that in control group on the 5th day post scalding ( P < 0.01). But there was no significant difference in the expression of Cyclin C between asiaticoside treatment group and control group ( P > 0.05). Conclusion Asiaticoside can enhance the expression of CyclinB1 and PCNA,make cell cycle S+G 2 ahead and accelerate wound tissue healing.
