胶原表面改性CPC对小鼠卵黄囊间质干细胞体外生长与BMP-2表达的影响 被引量:1

摘要 目的 观察胶原表面改性磷酸钙骨水泥 (CPC)对小鼠卵黄囊间质干细胞体外生长与骨形态发生蛋白 2 (BMP 2 )表达的影响。方法 分离、纯化及传代培养小鼠卵黄囊间质干细胞 ,用0 .5 %Ⅰ型胶原对CPC进行表面改性 ,检测胶原表面改性CPC对卵黄囊间质干细胞增殖分数的影响 ,扫描电镜观察卵黄囊间质干细胞在胶原表面改性CPC表面的黏附生长情况 ,逆转录 聚合酶链式反应 (RT PCR)检测与胶原表面改性CPC共培养卵黄囊间质干细胞BMP 2mRNA的表达。结果 胶原表面改性CPC有利于卵黄囊间质干细胞黏附生长 ,共培养 6d ,卵黄囊间质干细胞的增殖分数为 1.12 9,并可检测到BMP 2mRNA的表达。结论 胶原表面改性CPC能支持小鼠卵黄囊间质干细胞体外生长 ,并促进其BMP 2mRNA的表达。 Objective To investigate the effects of type Ⅰ collagen surface modified CPC on the growth and expression of BMP 2 of murine yolk sac mesenchymal stem cells (mYS MSCs).Methods mYS MSCs were separated from yolk sac and purified via passaged culture.To improve the properties of CPC,type Ⅰ collagen was used for surface modification of CPC.The proliferation index (PI) was used to evaluate the effects of ColⅠ/CPC on the proliferation of mYS MSCs.The adhesion of mYS MSCs on the surface of ColⅠ/CPC was observed under a scanning electron microscope.The mRNA expression of BMP 2 of mYS MSCs was detected by RT PCR.Results The PI of mYS MSCs was 0.811 after co cultured with CPC for 6 days,while that of mYS MSCs was increased to 1.129 ( P < 0.05) after co cultured with type Ⅰ collagen surface modified CPC.The mRNA expression of BMP 2 was detected in mYS MSCs after co cultured with ColⅠ/CPC for 6 days.Conclusion The adhesion,proliferation and the mRNA expression of BMP 2 of mYS MSCs were promoted by type Ⅰ collagen surface modified CPC.
