摘要
目的探讨血管紧张素Ⅱ(AngⅡ)对血管平滑肌细胞(VSMCs)基质金属蛋白酶2(MMP2)活性的影响及其与细胞增殖的关系。方法体外培养大鼠主动脉VSMCs,用AngⅡ进行刺激,采用明胶酶图法检测MMP2的活性。结果经AngⅡ处理12、24、48h后,VSMCs的MMP2活性分别为未处理组的237、196、158倍(P<005)。但AngⅡ对VSMCs增殖的促进作用在刺激3h时较明显(P<005)。结论AngⅡ提高VSMCs的MMP2活性晚于促进VSMCs增殖作用,显示AngⅡ这两种作用具有时相差异。
Objective To study the effect of angiotensinⅡ( AngⅡ) on the activity of matrix metalloproteinase-2 (MMP-2) in vascular smooth muscle cells (VSMCs) and its relationship with cell proliferation. Methods The aorta VSMCs of rats were cultured in vitro and stimulated with AngⅡ. The activity of MMP-2 was measured by gelatin zymography. Results The activities of MMP-2 of VSMCs treated by AngⅡfor 12, 24 and 48 hours were higher than those of the control by 2.37 , 1.96 and 1.58 times respectively (P<~0.05 ). But the promotive effect of AngⅡon the proliferation of VSMCs was obvious 3 hours after simulation (P<0.05 ). Conclusions There is phage difference between the effect of Ang Ⅱ on promoting cell proliferation and enhancing the activity of MMP-2 of VSMCs.
