高效液相色谱法测定消渴降糖胶囊中槲皮素的含量

Quercetin determination by high pressure liquid chromatography in xiaoke jiangtang capsules

目的建立消渴降糖胶囊中槲皮素含量测定的方法。方法采用高效液相色谱法,色谱柱为Kromasil-C18(4.6mm×250mm,粒径5μm),甲醇-0.2%磷酸溶液(48:52)为流动相,检测波长360nm,流速1.2ml/min,柱温40℃。结果槲皮素在26.24～131.2μg/ml范围内,浓度与峰面积呈线性关系(r=0.9989)。平均加样回收率为100.0%(RSD=3.10%)。结论该方法简单、准确、可靠、专属性强、重现性好,可作为消渴降糖胶囊的质量控制方法。

Objective To establish a method to determine the content of quercetin in xiaoke jiangtang capsules. Methods High pressure liquid chromatography (HPLC) was used with Kromasil-C18 column. The mobile phase consisted of methanol-0.2% phosphoric acid (48:52) with a flow rate of 1.2 ml/min, and the wavelength of detector was 360 nm. Results The linear range of quercetin was 26.24～131.2 μg/ml, and the average recovery was 100.0% (RSD=3.10%). Conclusion This method is simple, accurate, reliable with good specificity and reproducibility, it can be used as a quality control method for xiaoke jiangtang capsules.
