摘要 To prepare a hand-made micropore membrane culture plate insert forco-culture. Methods The plate insert was made using plastic centrifuge tube and micropore membrane.After seeding brain capillary endothelial cells (BCECs) on it (under the effect ofastrocyte-conditioned medium), the plate insert was assessed by analysis of trans-endothelialelectrical resistance (TEER). Results The plate insert has a stability of at least 15 d underculture condition. TEER increased significantly under co-culture condition from (66.1 +- 13.3)Ωcm^2 to (182.2 +- 6.7) Ωcm^2. Conclusion This micropore membrane culture plate insert can beeasily made, on which BCEC culture can be successfully performed. Moreover, it is adjustable andrecyclable. It follows that the plate insert is a useful tool for co-culture and the relatedresearch fields. 目的 探讨一种新型微孔底膜插入式培养皿的制作方法 ,为进行体外细胞共培养提供技术支持。方法 利用塑料离心管 ,微孔滤膜为材料制备微孔底膜插入式培养皿 ;在该插入式培养皿中接种大鼠脑微血管内皮细胞 (braincapillaryen dothelialcells ,BCECs) (在星形胶质细胞条件培养液影响下 ) ,以跨内皮细胞阻抗 (trans endothelialelectronicresistance ,TEER)为指标对该插入式培养皿进行评价。结果 该插入式培养皿在培养条件下 (未接种细胞 ) 1 5天内未出现破损 ;共培养条件下TEER增幅明显 :由 (6 6 1± 1 3 3)Ωcm2 升至 (1 82 2± 6 7)Ωcm2 。结论 该微孔底膜插入式培养皿制作简便 ,脑微血管内皮细胞在上生长良好 ,并具有可调节性和可重复使用的特点 ,是一种可用于体外细胞共培养相关研究工作有效的工具。
