摘要 利用RAPD标记研究了 2 9个烤烟品种的分子指纹和遗传关系。 10 0个随机引物经 10个品种筛选 ,选出 18个随机引物对 2 9个品种基因组DNA进行扩增 ,得到了 2 9个烤烟品种的分子指纹图谱 ,参照这些图谱 ,可对不同烤烟品种进行真实性鉴定。以扩增到的 79条多态性DNA片段为基础计算 2 9个品种间遗传相似系数 ,利用UPGMA法作聚类图 ,可将参试品种分为 3个类群。云南省主栽品种K3 2 6与近年选育的品种V2、云烟 85、云烟 3 17等被归为一类 ,遗传基础过于狭窄 ,建议选用一些遗传关系较远的品种进行亲本选配、品种布局 ;北方烟区主栽品种NC89。 The molecular fingerprints and genetic relationships among 29 flue cured tobacco varieties were studied using RAPD markers. By pre screening of 100 random decamer primers,18 primers were applied to RAPD analysis. All 29 varieties had their own band patterns or molecular fingerprints. RAPD technology can be used to identify different flue cured tobacco varieties. Based on the polymorphic hands amplified by RAPD, the genetic similarities between varieties were calculated. The dendrogram constructed using UPGMA can classify these varieties into 3 groups. K326, the widely cultivated cultivar, and the varieties developed recently in Yunnan, such as V2, Yunyan 85,Yunyan 317,were grouped together. It showed the low genetic diversity among the commercial cultivars in Yunnan. Selecting the varieties having distant genetic relationships in the selection of the parents and the distribution of the cultivars was suggested. NC89, Jingyehuang, and G140 was clustered in another group.
