摘要
目的 比较研究古尼虫草纯多糖组分FP及其解聚物的免疫活性。方法 采用超声波解聚多糖 ,并用葡聚糖凝胶分离获得解聚级分 ,通过MTT法和中性红比色法测定其对小鼠脾淋巴细胞增殖、腹腔巨噬细胞吞噬功能和细胞毒T淋巴细胞杀伤功能的影响。结果 FP的解聚级分FP 1、FP 2和FP 3有更强的免疫活性 ,在中、低剂量 (1和 10 μg mL)与对照相比有显著差异 ,而其中以FP 2的活性最明显。结论 古尼虫草多糖解聚后能明显提高免疫活性 。
Objective To investigate the immunocompetence changes after pure polysaccharide (FP) was depolymerized by ultrasonication in Cordyceps gunnii. Methods The FP was depolymerized by ultrasonication and isolated by a Sephadex A-25 column. The effects of FP and its depolymerizing substance on mouse spleen lymphocytes proliferation, mouse peritoneal macrophage (PMФ) phagocytosis, and cytotoxin T lymphocytes (CTL) activity were studied by MTT method, neutral red colorimetry, respectively. Results The depolymerizing fractions of FP-1, FP-2, and FP-3, especially the FP2, had higher immunocompetence than FP did. Compared with control group, three kinds of depolymerizing fractions all had evident differences at doses of 1 μg/mL and 10 μg/mL.Conclusion Depolymerizing substance of pure polysaccharide from C. gunnii has higher immunocompetence.
